Narration : 11. Centrifugation
- First, prepare the sample in the centrifuge tubes. Keep the sample volume below the half, or the liquid might overflow upon centrifugation.
- Along with the sample tube, another tube is required to maintain the balance. If the weights of the two tubes are not equal, a serious accident may occur.
- On one side of the balance, place the centrifuge tube containing the sample and in the other place the tube for balance.
- Add distilled water little by little into the balance tube until its weight becomes equal to that of the sample tube.
- Insert the pair of tubes in two holders located at opposite positions in the rotor. Close the cover tightly.
- Start centrifugation by slowly turning the speed dial to a preset value.
- The spinning of high-speed results in centrifugal force and separates the precipitate.
- It is strictly forbidden to open the cover or put your hand into the machine.
- Turn off the machine and wait for the rotor to stop completely.
- Then, take out the tubes carefully not to disturb the precipitate.
